两台功能强大的X射线仪器——直线加速器相干光源(Linac Coherent Light Source,LCLS)和斯坦福同步辐射光源(Stanford Synchrotron Radiation Lightsource,SSRL)正在帮助科学家深入了解诸如昏睡病这样的疾病,并可能由此找到新的治疗方法。

这两台仪器都设在美国能源部(U.S. Energy Department)的一间实验室中。据能源部介绍,直线加速器相干光源产生的X射线脉冲强度极高,能够以单个原子的分辨率来显示物质结构。这一敏度最近帮助解析了与昏睡病传播有关的一种酶的结构。

研究人员在活体昆虫细胞中培养了一种昏睡病蛋白的微小晶体,并将它们串流在X射线激光中。通过解析来自这些晶体的衍射图案的光的不同强度的图谱,如上图所示,科学家希望能够制作出这种酶的结构的模型。

非洲昏睡病寄生虫用以加速其感染的蛋白质与一种人类蛋白质相当类似。一份足够精确的结构图谱可以显示两者之间的一些本质上的差异,从而使研究人员能够研发出以寄生虫蛋白质为对象同时又不触及人类蛋白质的药物。

昏睡病由一种原虫引起,通过采采蝇感染人体,它威胁着非洲数百万人,估计每年有30,000人死于此病。

而斯坦福同步辐射光源则被研究人员用来制作使幽门螺旋杆菌中和胃酸的一种结构的三维图像。幽门螺旋杆菌可引发胃溃疡并可增加患胃癌的机率。

Two powerful X-ray instruments, the Linac Coherent Light Source and the Stanford Synchrotron Radiation Lightsource, are helping scientists learn more about diseases like sleeping sickness and could lead to new treatments.

Two powerful X-ray instruments, the Linac Coherent Light Source (LCLS) and the Stanford Synchrotron Radiation Lightsource (SSRL), are helping scientists learn more about diseases and could lead to new treatments.

According to the U.S. Energy Department, which hosts both instruments at one of its labs, the LCLS produces X-ray pulses so powerful that they can show structures down to the resolution of individual atoms. This sensitivity recently helped reveal the structure of an enzyme involved in the spread of sleeping sickness.

Researchers grew tiny crystals of a sleeping sickness protein in live insect cells and streamed them into the path of the X-ray laser. By mapping different intensities of light from the crystals’ diffraction patterns, shown above, scientists hope to be able to model the enzyme’s structure.

The protein used by the African sleeping sickness parasite to speed its infection is quite similar to a human protein. A precise-enough map of structures could show some essential differences between the two and enable researchers to design drugs targeting the parasite’s protein while leaving the human version untouched.

Sleeping sickness, caused by a protozoan that infects people via tsetse flies, threatens millions across Africa and kills an estimated 30,000 people every year.

The SSRL has been used by researchers to produce a 3-D image of a structure that enables the bacterium H. pylori to neutralize stomach acid. H. pylori causes stomach ulcers and can increase one’s odds of developing stomach cancer.
