office确实有“办公室”的意思
但是in office 中的office表示【官职；公职】 比如：hold office 政府任职the office of vice president 副总统职位in office则表示 在职；执政；在政府担任要职
例句：

How long has he been in office ? 

他任职多久了？

那问题来了，“我在办公室”到底该咋说？↓↓↓最贴近我们说的“我在办公室”应该是：I'm in the office.例句：I'm in the office now. Please help me pick up my delivery downstairs.我在办公室，请帮我去楼下取下外卖。
