抗荧光淬灭剂Fluorescent Mounting Media(71-00-06)

    荧光分子与溶剂或其他分子之间相互作用，使荧光强度减弱的现象称为荧光猝灭。这通常是因为荧光分子在激发光的照射下，反复经历多次激发，发射过程后，往往造成分子内部结构发生不可逆的变化，不能吸收更多的光子而进一步发射荧光。这时就称该分子被光漂白了。当激光功率高时，荧光分子有可能在高强度的激光下被漂白了。引起荧光强度降低的物质称为淬灭剂。当荧光物质浓度过大时，会产生自淬灭现象。

    KPL公司针对荧光试验中遇到的光漂白或者荧光淬灭现象，专门开发出的Fluorescent Mounting Media是抵抗荧光信号淬灭的试剂。其主要原理是可以有效避免荧光染色后的固定细胞或组织等样品不被氧化，从而防止荧光淬灭导致的荧光信号减弱现象。

KPL的抗荧光淬灭剂（Fluorescent Mounting Media）是一种基于甘油溶液，并包括用于减缓各种荧光（如绿色FITC荧光、红色Rhodamine荧光等）淬灭的封片试剂，该试剂不包括p-苯二胺及乙醇组分，可以用于减缓各种常见荧光染料的荧光淬灭。我们推荐在免疫组化实验、免疫细胞荧光以及荧光原位杂交等实验中使用该抗荧光淬灭剂。 

    KPL的荧光抗淬灭剂操作简单，当组织块或细胞样本与荧光标记的试剂结合反应，并充分冲洗后，在封片前用滴加一滴抗荧光淬灭封片液，盖上盖玻片就可以了。该抗荧光淬灭剂还能用于荧光抗体或其它耦联物的溶解，通常用等体积的抗荧光淬灭剂和试剂纯级的水溶液溶解荧光抗体，可大大提高荧光抗体的工作效果。

DESCRIPTION

   Fluorescent Mounting Media is a glycerol-based solution containing an inhibitor that retards photobleaching of fluorochromes. It is not a permanent mounting medium. It does not contain p-phenylenediamine or alcohol.

FORM/STORAGE

    Fluorescent Mounting Media is provided as a ready to use liquid. Store tightly capped at room temperature. Stable for a minimum of one year from date of receipt when stored at room temperature.

APPLICATIONS

    Fluorescent Mounting Media is recommended for immunohistochemistry and fluorescent in situ hybridization.

USE

    After tissue sections or other cellular preparations have been reacted with fluorochrome-labeled reagents and washed thoroughly, place a drop of Fluorescent Mounting Media on specimen. Coverslip and evaluate as soon as possible.

For rehydration of lyophilized fluorochrome conjugates, the use of a solution consisting of 50% Mounting Media and 50% reagent quality water may increase the working titer of the antibody.

The use of buffered saline solution to dilute the Mounting Media is not recommended.
